How to set up a vial – extraction by EME

１　Membrane attached to union

Attach a circular membrane to the union. There is a doughnut-shaped insulating stopper on the inner wall of the union midsection. Press the membrane into the unit while tapping it lightly so that the outer edge of the membrane is firmly in contact with the insulating stopper to the extent that the pipette tip will not tear the membrane.

2 Injection of solution into vials

Stand the donor and acceptor vials in a commercially available vial holder and fill them with sample solution and acceptor solution, respectively.

３　Union attachment to acceptor vial

Attach the union with the membrane installed to the acceptor vial. Screw it in straight and tight to prevent leakage. At this time, install the membrane so that the fully visible side of the membrane is in contact with the acceptor vial.

4 Constructing SLM

Drop the required volume (usually about 10 μl) of the optimal organic solvent onto the membrane installed in the union and allow it to infiltrate the membrane to construct the SLM (Supported Liquid Membrane). Reference extraction conditions for target compounds can be found at how-to-eme.com. Enter the compound name in English in the search window and press “Enter”; extraction conditions will be displayed only if the compound can be extracted by EME. If the recovery rate is low, you will need to optimize the extraction conditions for each sample and target compound.

5　Connection of acceptor vial to donor vial

Connect the donor vial to the other side of the union with the acceptor vial attached (the side where the insulating stopper of the union is visible).

6 Loading the vial unit into the vial holder

Load the vial unit into the vial holder of the ETN-12 EME paying attention to the voltage direction (extraction direction).

7　Parameter setting and EME start

Set various parameters (voltage, running time, and stirring rpm intensity) while checking the instruction manual, and start EME.
